I have idiopathic peripheral neuropathy ... the only thing they found that would work is lyrica. I picked up some CBD oil yesterday morning. I am prescribed to take 75 mg of lyrica 3x per day. I took one yesterday morning and have only used the CBD oil since. I bought the Koi brand, flavored, 250 MG. I used a full dropper yesterday late morning and a full dropper yesterday late afternoon. I used it once today (one full dropper) and I am amazingly pain free.
From this study we can conclude that the acute effects of THC (e.g. increased anxiety) are unfavorable. Evidence suggests that CBD appears well-tolerated and safe, with no adverse physiological reactions compared to a placebo. However, since the physiological effects of CBD (600 mg) were of no statistically significant difference from the placebo, it is unclear if CBD elicits any therapeutic effect – even at a seemingly reasonable dose.
Cannabidiol (CBD) is a component of Cannabis sativa that has a broad spectrum of potential therapeutic effects in neuropsychiatric and other disorders. However, few studies have investigated the possible interference of CBD on the sleep-wake cycle. The aim of the present study was to evaluate the effect of a clinically anxiolytic dose of CBD on the sleep-wake cycle of healthy subjects in a crossover, double-blind design. Twenty-seven healthy volunteers that fulfilled the eligibility criteria were selected and allocated to receive either CBD (300 mg) or placebo in the first night in a double-blind randomized design (one volunteer withdrew from the study). In the second night, the same procedure was performed using the substance that had not been administered in the previous occasion. CBD or placebo were administered 30 min before the start of polysomnography recordings that lasted 8 h. Cognitive and subjective measures were performed immediately after polysomnography to assess possible residual effects of CBD. The drug did not induce any significant effect (p > 0.05). Different from anxiolytic and antidepressant drugs such as benzodiazepines and selective serotonin reuptake inhibitors, acute administration of an anxiolytic dose of CBD does not seem to interfere with the sleep cycle of healthy volunteers. The present findings support the proposal that CBD do not alter normal sleep architecture. Future studies should address the effects of CBD on the sleep-wake cycle of patient populations as well as in clinical trials with larger samples and chronic use of different doses of CBD. Such studies are desirable and opportune.

While there are more unknowns than knowns at this point, Grant says he doesn’t discount all the anecdotal CBD reports. “You hear somebody say, ‘Hey, I gave this to myself and my kid and we feel a lot better,’ and we should never dismiss that kind of information,” he says. Participants were recruited through advertisements in the local media of the city of Ribeirão Preto, São Paulo, Brazil. Initially, 335 individuals who were interested in participating were evaluated, 265 of whom were excluded in the recruitment interview (which contained questions about clinical data, demographics, psychiatric symptoms, sleep patterns, among others). The remaining 70 participants were asked to keep a sleep log and completed the rating scales on sleep patterns (ESS, Epworth Sleepiness Scale; PSQI, Pittsburgh Sleep Quality Index). After these procedures, 27 participants were considered eligible for the study (Figure Figure11) and were randomized into two groups (group 1: placebo – CBD, group 2: CBD – placebo) matched in terms of sex, age, and years of education. To ensure the adequacy of the matching procedure, one participant of each pair had his treatment blindly chosen between the two treatment options available and the other participant (matched to the first one) was assigned to the remaining option.
The relative representativeness of the small sample size and the use of a single dose of CBD can perhaps be regarded as a limitation of our study, as it does not allow the assessment of the effects of chronic treatment with CBD on sleep. In the study by Chagas et al. (2014b), for example, CBD was chronically administered for 6 weeks to patients with Parkinson’s disease and REM sleep behavior disorder. Since the effects of CBD are biphasic (Zuardi et al., 2017), the use of a single dose also limits the interpretation of the present findings. Moreover, monitoring changes in sleep using a conventional polysomnography presents some intrinsic limitations, as it is insufficient alone to detect drug-induced changes of the sleep EEG. For this purpose, a spectral analysis or a similar procedure is also needed. Conversely, the use of preclinical polysomnography to characterize drug-induced sleep disturbances has been increasingly recommended in the regulatory context (Authier et al., 2016). Finally, it is essential to evaluate the effects of CBD in a larger sample and in individuals diagnosed with sleep disorders in addition to healthy volunteers.
I have been totally off the effexor and all anti-depressants for 2 weeks now. The dizziness is getting much better however my emotions/agitation are horrible. I cry at everything and am extremely crabby/agitated. I realize most of this has to do with the withdrawal. I really want to see this through to find out if I can live without anti-depressants but at the same time I know it's very hard on my family. I have another doctor appt beginning of April and she says that if I don't feel better by then I most likely will need to go back on an anti-depressant. For the most part I agree with her. My hopes of proving her wrong as getting slim however. I'd like to know how long it took some of you who have withdrawn from anti-depressants to feel somewhat 'normal' or you knew you had to go back on them? I guess I'm asking if another month is a good amount of time for me to determine what I should do. In some ways I feel like I should start on them again now but I'm not going there yet? BTW, I am in no way feeling suicidal. Mornings seem to be my worst time and by early evenings I feel somewhat better – is this strange too? I haven't tried the CBD living water yet but did find a place near me to get it. Just havent had the time to get there. I also have the Ativan which I take one night to help with sleep. I'm trying not to take it unless really necessary. Tomorrow I have a huge even that my husband and I are in charge of so I'm planning to take an Ativan in the morning to get me through the day without falling apart (crying scene) in front of everyone (or yelling at them) :)! Following cloning of the endogenous receptor for THC, namely the CB1R, endogenous CB1R ligands, or “endocannabinoids” (eCBs) were discovered, namely anandamide (AEA) and 2-arachidonoylglycerol (reviewed in ). The CB1R is an inhibitory Gi/o protein-coupled receptor that is mainly localized to nerve terminals, and is expressed on both γ-aminobutryic acid-ergic and glutamatergic neurons. eCBs are fatty acid derivatives that are synthesized on demand in response to neuronal depolarization and Ca2+ influx, via cleavage of membrane phospholipids. The primary mechanism by which eCBs regulate synaptic function is retrograde signaling, wherein eCBs produced by depolarization of the postsynaptic neuron activate presynaptic CB1Rs, leading to inhibition of neurotransmitter release . The “eCB system” includes AEA and 2-arachidonoylglycerol; their respective degradative enzymes fatty acid amide hydroxylase (FAAH) and monoacylglycerol lipase; the CB1R and related CB2 receptor (the latter expressed mainly in the periphery); as well as several other receptors activated by eCBs, including the TRPV1 receptor, peroxisome proliferator-activated receptor-γ, and G protein-coupled 55 receptor, which functionally interact with CB1R signaling (reviewed in [21, 24]). Interactions with the TRPV1 receptor, in particular, appear to be critical in regulating the extent to which eCB release leads to inhibition or facilitation of presynaptic neurotransmitter release . The TRPV1 receptor is a postsynaptic cation channel that underlies sensation of noxious heat in the periphery, with capsacin (hot chili) as an exogenous ligand. TRPV1 receptors are also expressed in the brain, including the amygdala, periaqueductal grey, hippocampus, and other areas [26, 27].

Few interactions: Most evidence indicates that CBD is unlikely to interact with pharmaceutical drugs. After arrival at the Clinical Research Unit of the Ribeirão Preto Medical School University Hospital (Ribeirão Preto, Brazil) written informed consent was signed, the subjective measures (STAI, VAMS) of the participants were collected and the electrodes used for the polysomnography exam were placed. Next, the subjective measures were completed once again (STAI, VAMS) and, 30 min before the beginning of the polysomnographic examination, the single dose of CBD (300 mg) or placebo was administered. Polysomnography recordings were performed over 8 h. On the morning after the examination, the electrodes were removed from the subject and the VAMS, STAI, WAIS, and PVT were completed. I’m 48 and have been diagnosed with anxiety disorder, depression, ptsd and have had panic attacks that have lead me to the ER 3 or 4 times. My psychiatrist put me on wellbutrin and klonopin for the anxiety and depression… I’m taking very low dosages of each but from what I’ve read when you come off of the klonopin it has physical side effects. I’m wanting to come off of both and my psychiatrist doesn’t think good things about cannabis and says that it interferes with the GABA receptors in the brain. I’m trying to find a doctor than can explain to me face to face how CBD and THC work on the brain and what he/she would recommend I do to get off the big pharma train. I’m in Puerto Rico.

At lower doses, CDB (15 mg/day) co-administered with tetrahydrocannabinol (THC, 15 mg/day) increased wakefulness (Nicholson et al., 2004). More recently, Chagas et al. (2014b) investigated the effects of chronically administered CBD (75–300 mg per day for 6 weeks) in patients with Parkinson’s disease and found a reduction in symptoms of REM sleep behavior disorder. After discontinuation of the drug, the frequency of symptoms returned to baseline levels, prior to treatment with CBD. Overall, existing preclinical evidence strongly supports the potential of CBD as a treatment for anxiety disorders. CBD exhibits a broad range of actions, relevant to multiple symptom domains, including anxiolytic, panicolytic, and anticompulsive actions, as well as a decrease in autonomic arousal, a decrease in conditioned fear expression, enhancement of fear extinction, reconsolidation blockade, and prevention of the long-term anxiogenic effects of stress. Activation of 5-HT1ARs appears to mediate anxiolytic and panicolytic effects, in addition to reducing conditioned fear expression, although CB1R activation may play a limited role. By contrast, CB1R activation appears to mediate CBD’s anticompulsive effects, enhancement of fear extinction, reconsolidation blockade, and capacity to prevent the long-term anxiogenic consequences of stress, with involvement of hippocampal neurogenesis.
Dr. Robert Carson is a pediatric neurologist at Vanderbilt University who has evaluated the effectiveness of CBD supplements in kids with seizures. He says the supplements can be beneficial for these children. However, he says, if the FDA follows its advisory panel's advice and approves a pharmaceutical-grade CBD drug, that would open up a new treatment option by delivering a high-quality, consistent dose of CBD.
