In 1992 Mechoulam’s quest for quantification led him from the plant itself to the inner recesses of the human brain. That year he and several colleagues made an extraordinary discovery. Hi Celeste. Thanks for your question. I would say as long as you feel comfortable with it, you can increase the dose for sleep to see if it has a stronger effect on your insomnia. You can carefully increase the dosage by another half or full dropper-full and see if that helps. In regard to how much to take during the day, how much are you currently using during the day?

My favorite thing about it is how incredibly mild it is – like I said, the effects just kind of slowly ooze their way in without you even really noticing. Also, I love how seemingly long-lasting the effects are. I’ve read that some people prefer vaping over taking the oil drops because they say vaping is more potent, but I also understand that the effects of vaping are much shorter lived.
Stephanie, generally, I have patients take 20 to 150mg a day for sleep +/- anxiety. Start low and go slow. Know the dosages of your product. Usually 2/3 to 3/4 of the daily dose is 1-2 hours before bedtime, and the other portion is upon waking (to improve wakefulness during the day). Other factors such as stress, hormone replacement, other meds & medical conditions, etc. play a role along with individual differences. I own a compounding pharmacy, so we see a lot of unique needs. I can't give more specific advice in this forum, but there is help!
Following cloning of the endogenous receptor for THC, namely the CB1R, endogenous CB1R ligands, or “endocannabinoids” (eCBs) were discovered, namely anandamide (AEA) and 2-arachidonoylglycerol (reviewed in [22]). The CB1R is an inhibitory Gi/o protein-coupled receptor that is mainly localized to nerve terminals, and is expressed on both γ-aminobutryic acid-ergic and glutamatergic neurons. eCBs are fatty acid derivatives that are synthesized on demand in response to neuronal depolarization and Ca2+ influx, via cleavage of membrane phospholipids. The primary mechanism by which eCBs regulate synaptic function is retrograde signaling, wherein eCBs produced by depolarization of the postsynaptic neuron activate presynaptic CB1Rs, leading to inhibition of neurotransmitter release [23]. In addition to dose-dependent activation of TRPV1 channels, the anxiogenic versus anxiolytic balance of CB1R agonists also depends on dynamic factors, including environmental stressors [33, 49].
