Research has demonstrated the effect of cannabidiol as a 5-HT1A receptor partial agonist, meaning it binds to the receptor site but only stimulates the receptor partially (relative to a full agonist). Studies with cloned human cell cultures note that cannabidiol displaces 5-HT1A agonists from 5-HT1A receptor sites in a dose-dependent manner.
The extract known as CBD oil sold in the U.S. falls into one of two categories. Crystalline isolate exclusively contains CBD, as other cannabinoids have been removed; full spectrum oil, on the other hand, retains THC and other cannabinoids, and is only sold in states where marijuana use has been legalized. CBD oil can be consumed several different ways, including ingested capsules and food products, vaporizing, tinctures, and topical creams. To determine the effects of each substance, physiological measures were collected along with symptom ratings approximately 1, 2, and 3 hours post-administration. Post-trial assessment of physiological measures indicated that compared to placebo and CBD, administration of THC caused anxiety, dysphoria, positive psychotic symptoms, neurophysiological sedation, and elevated heart rate. Strikingly, there appeared to be no significant differences between CBD and placebo on physiological or symptomatic measures.
The Green roads CBD didn’t work for me at all. The CBD is mixed with glycerine & it doesn’t come out of the dropper well at all so it’s hard to know how much you are getting. Plus I think glycerine is not a good carrier at all. A simple coconut oil mix would work much better for absorption. I think there are much better options for your money! Really disappointed:(

Participants were recruited through advertisements in the local media of the city of Ribeirão Preto, São Paulo, Brazil. Initially, 335 individuals who were interested in participating were evaluated, 265 of whom were excluded in the recruitment interview (which contained questions about clinical data, demographics, psychiatric symptoms, sleep patterns, among others). The remaining 70 participants were asked to keep a sleep log and completed the rating scales on sleep patterns (ESS, Epworth Sleepiness Scale; PSQI, Pittsburgh Sleep Quality Index). After these procedures, 27 participants were considered eligible for the study (Figure Figure11) and were randomized into two groups (group 1: placebo – CBD, group 2: CBD – placebo) matched in terms of sex, age, and years of education. To ensure the adequacy of the matching procedure, one participant of each pair had his treatment blindly chosen between the two treatment options available and the other participant (matched to the first one) was assigned to the remaining option.
Given the current state of evidence, it is important to avoid assuming CBD is a sustainable long-term intervention for anxiety disorders. As further research is conducted with larger-scale, longer-term, robustly designed trials – we will better understand the anxiolytic capacity of CBD. Those with refractory cases of anxiety in search of an alternative pharmacological intervention may want to discuss the feasibility of “as-needed” CBD administration with a medical professional.
Although nearly all of the published studies found CBD effective for the attenuation of anxiety, there are some notable limitations associated with the research. Perhaps the most notable limitation is the fact that most CBD studies investigate the effect of acute, single-dose administration. The problem with this is that it remains unclear as to whether chronic or long-term CBD ingestion maintains therapeutic efficacy.
Although not as abundant as THC cannabinoid content, cannabidiol accounts for approximately 40% of all cannabinoids within cannabis extract. Unlike THC, cannabidiol is non-psychoactive and isn’t typically ingested with the intent to attain any sort of psychological euphoria. That said, the medicinal properties associated with cannabidiol (often administered in the format of “CBD oil”) are thought to far exceed those of THC.
Anxiolytic effects of CBD in models of generalized anxiety have been linked to specific receptor mechanisms and brain regions. The midbrain dorsal periaqueductal gray (DPAG) is integral to anxiety, orchestrating autonomic and behavioral responses to threat , and DPAG stimulation in humans produces feelings of intense distress and dread . Microinjection of CBD into the DPAG produced anxiolytic effects in the EPM, VGC, and ETM that were partially mediated by activation of 5-HT1ARs but not by CB1Rs [65, 68]. The bed nucleus of the stria terminalis (BNST) serves as a principal output structure of the amygdaloid complex to coordinate sustained fear responses, relevant to anxiety . Anxiolytic effects of CBD in the EPM and VCT occurred upon microinjection into the BNST, where they depended on 5-HT1AR activation , and also upon microinjection into the central nucleus of the amygdala . In the prelimbic cortex, which drives expression of fear responses via connections with the amygdala , CBD had more complex effects: in unstressed rats, CBD was anxiogenic in the EPM, partially via 5-HT1AR receptor activation; however, following acute restraint stress, CBD was anxiolytic . Finally, the anxiolytic effects of systemic CBD partially depended on GABAA receptor activation in the EPM model but not in the VCT model [61, 62].
Throughout the SPST, researchers utilized the Visual Analogue Mood Scale (VAMS), Negative Self-Statement scale (SSPS-N), and physiological measures (blood pressure, heart rate, skin conductance) to determine the anxiolytic efficacy of CBD. Results indicated that those receiving the CBD exhibited significantly less anxiety, cognitive deficits, speech discomfort, and decreased alertness on the VAMS measure. Contrastingly, the individuals receiving the placebo exhibited high anxiety, cognitive deficits, high alertness, and overall discomfort on the VAMS.
CBD was first discovered in the 1940s by Roger Adams, the former head of the chemistry department at University of Illinois at Champaign-Urbana. In his research, Adams isolated CBD from hemp but couldn’t determine what exactly he’d found. In addition to CBD, Adams also synthesized analogs of THC and another cannabinoid, showing their relationship to CBD.

A 2007 review published in the journal JAMA Internal Medicine found that smoking marijuana resulted in similar declines in respiratory system health as smoking tobacco.
